MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA type-c) is a 16-amino acid peptide encoded within the 12S ribosomal RNA gene of the mitochondrial genome — making it one of a novel class of mitochondria-derived peptides (MDPs). First described in 2015, MOTS-c acts as a mitochondrial stress sensor and systemic signalling molecule: under metabolic stress, it translocates from mitochondria to the nucleus, where it activates AMPK and regulates metabolic gene expression.
