MOTS-c

Overview

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a 16-amino acid peptide encoded within the mitochondrial genome — one of very few peptides with a mitochondrial rather than nuclear origin. Discovered in 2015, it represents a new class of signalling molecule called mitochondrial-derived peptides (MDPs).

Research Context

MOTS-c functions as a metabolic regulator with activity analogous to exercise. Research has demonstrated that it translocates to the nucleus under metabolic stress and activates AMPK pathways, improving glucose uptake and insulin sensitivity independent of insulin signalling. Animal studies have shown reversal of diet-induced obesity, improved skeletal muscle function, and lifespan extension in aged mice. It also modulates the folate cycle and one-carbon metabolism. Human circulating levels of MOTS-c decline with age and correlate inversely with metabolic disease markers, making it a subject of significant interest in longevity and metabolic health research.

MOTS-c requires intestinal protection from proteolysis before reaching systemic circulation.
