Semax and Selank were both developed at Soviet and Russian state research institutes in the 1980s and 90s and have decades of clinical use in Russian neurology. They share nasal delivery and BBB penetration, but hit entirely different receptor targets — making them complementary rather than redundant.
Why these two compounds
Semax is a synthetic analogue of ACTH(4–10), the fragment of adrenocorticotropic hormone responsible for cognitive function. It upregulates BDNF mRNA expression in the hippocampus — activating the gene that produces Brain-Derived Neurotrophic Factor, the protein responsible for synaptic plasticity and neural connection maintenance. It crosses the blood-brain barrier in under 4 minutes via the olfactory epithelium, with a single dose active for up to 24 hours. It does not raise cortisol or act as a stimulant. Its mechanism is gene-level.
Selank is a synthetic analogue of the endogenous immunomodulatory peptide tuftsin, developed at the Institute of Molecular Genetics. Its primary mechanism is GABA-A receptor modulation, producing an anxiolytic effect without sedation or dependence. It also has documented immunomodulatory activity. Where Semax sharpens cognitive performance, Selank reduces stress-related interference with it — the two effects are additive in practice.
The pairing addresses both sides of cognitive performance: the neural substrate (BDNF, synaptic plasticity) via Semax, and the stress interference layer (GABA, cortisol modulation) via Selank.
Protocol reference
Research protocol: Semax 200–600mcg intranasal once or twice daily; Selank 250–500mcg intranasal once or twice daily. Semax is typically run in the morning for focus; Selank can be run any time anxiety or stress load is the primary concern. Sniff after administration — tilting the head back reduces olfactory epithelium contact and bioavailability.
